- ItemEmbargoEfeitos da amitriptilina sobre a fibrose em modelo de obstrução vesical parcial(Universidade Federal de São Paulo (UNIFESP), 2010-04-28) Almeida Prado, Patricia Shirley de [UNIFESP]; Teixeira, Vicente de Paulo Castro [UNIFESP]; Universidade Federal de São Paulo (UNIFESP)Purpose: Chronic bladder outlet obstruction secondary to benign prostatic hypertrophy can reach up to 80% of men over 50 years and cause serious and irreversible changes throughout the urinary tract, especially in the bladder, however, current knowledge, therapeutics are still limited. Amitriptyline has recently been described as analgesic, anti-inflamatory and myorelaxing. Objective: To investigate the effects of amitriptyline hydrochloride on the process of fibrosis in a model bladder outlet obstruction in rats. Materials and Method: Wistar rats were subjected to infravesical obstruction and studied at intervals of 1 and 14 days after procedure. Animals were divided into 5 groups: sham, OIV1-T, OIV1-NT, OIV14-T e OIV14-NT. Bladder tissue was processed for histopathology, immunohistochemical and RT-PCR. Results: Groups OIV14 showed bladder fibrosis, hypertrophy of the smooth muscle cells and thickening of the bladder wall. Group OIV14-T demonstrated a higher intensity of fibrosis and higher macrophage infiltration rate and gene expression of TGF-β1. Conclusion: Amitriptyline presented an amplifying effect on the process of fibrosis as a whole in this experimental model.
