This study evaluated the effect of peptide 4 derived from the amino acid sequence of Lopap (Lonomia obliqua prothrombin activator protease) in culture of fibroblasts to evaluate the expression of molecules of extracellular matrix and the membrane receptors of interleukin-8 and in experimental model of skin lesion in rats to examine the effect of the molecule in the process of tissue repair. We used 40 male Wistar rats with 6 to 8 weeks, randomized to 5 experimental groups. After anesthesia, the rats were submitted to surgery, where eight circular full-thickness of skin from 6 mm of diameter were surrounded in the back and treated with saline or with peptide 4. After different periods (days 3, 7, 14 and 21 days after wounding) were carried out tests to evaluate the process of tissue repair between the groups. We conclude that peptide 4 is able to stimulate fibroblasts in culture and promote the acceleration of the process of tissue repair in vivo model with greater deposition of collagen, glycosaminoglycans and better tissue reorganization.
- ItemAcesso aberto (Open Access)Experimental model of the morphological and morphometric aspects of tissue repair in skin wounds submitted to beta radiation emitted by Strontium -90 in rats(Sociedade Brasileira para o Desenvolvimento da Pesquisa em Cirurgia, 2004-12-01) Pereira Filho, Gerson Vilhena [UNIFESP]; Camelo-nunes, José Mario; Yui, Fabrício; Universidade Federal de São Paulo (UNIFESP); ABC Medical SchoolThe purperose of this work is tell the use of the experimental model to investigation of the effects of Beta radiation of estroncium-90 for repairing the tissue of wounds made in rats. 48 animals of the lineage EPM-1 Wistar were used, distributed in two groups that received radiation in alternate days (group A) and daily (group B). Each group was divided in four groups of six rats to be analysed in the 3rd, 7th, 14th and 21st day after operation. Two incisions were made in the back of the animal and sutured immediately afterwards; the upper part was irradiated and the lower part used as control. On the dates established the irradiated and control wounds were macroscopically examined and withdrawn for preparation of the histological comparative study in the optical microscope. Following, an morphometrical analysis was performed to count leucocyts, fiberblast and colagen fibers which were submitted to statistical study.
