In the body, leptin has numerous functions in different tissues. In hematopoiesis, previous studies have demonstrated its potential in the differentiation and/or cell proliferation. The group of Dr. Antonio de Miranda, from Department of Biophysics of UNIFESP, has developed human and murine leptin synthetic peptide fragments, whose sequences included portions of the a helical region of the leptin molecule. Two of these human sequence peptides, Ac-hLEP92-11S-NH2and Ac-[Ser117]-hLEP116_140- NH2 have shown activity in a similar manner to the entire protein. Additionally, similar results were obtained with another homologous peptide sequence of mouse, the Ac-mLEP98-122-NH2.This study aimed to evaluate the modulation of hematopoiesis by human and murine synthetic peptides (based on sequences previously tested). The human, Ac-[Ser117]-hLEP116_140-NH2(LEP5), and murine, Ac-mLEP110-119-NH2(LEPF), sequence induced an increase of hematopoietic stem cell (HSC) and clonogenic capacity, as well as, activated Jak2 in HSC. Moreover, the fragment LEPF improved the engraftment of HSC after radiation, however did not accelerate bone marrow regeneration after the chemotherapy with 5-FU. The fragment LEPF only increased HSC after 5-FU treatment. Thus, the bioactive synthetic fragments of leptin could be used to improve the engraftment of HSC is some clinical situation such as HSC transplantation.
- ItemAcesso aberto (Open Access)Avaliação histológica, da apoptose e da proliferação no endométrio de mulheres com síndrome dos ovários policísticos(Universidade Federal de São Paulo (UNIFESP), 2013-11-27) Giordano, Luiz Augusto [UNIFESP]; Soares Junior, Jose Maria [UNIFESP]; Universidade Federal de São Paulo (UNIFESP)Introduction: The Polycystic Ovary Syndrome (PCOS) is a common endocrinopathy in women and can determinate infertility and endometrial diseases. The study of endometrial proliferation and apoptosis in these patients is still unclear. Objective: Evaluate the proliferation of endometrial cells by Ki67 and endometrial apoptosis by cleaved caspase-3, Bcl-2, Fas and FasL in patients with polycystic ovary syndrome compared with control patients, with normal cycles. Patients and methods: A cross-sectional and case-control study, including patients with polycystic ovary syndrome (PCOS) diagnosed using the criteria of the Androgen Excess Society Polycystic Ovary Syndrome (AE - PCOS , 2006) . We analysed 11 patients with PCOS, called GSOP, and eight patients in the control group , named GC . We analyzed clinical data ( menstrual cycles and body mass index ), metabolic data ( fasting glucose , total cholesterol and fractions , triglycerides ) , hormonal data ( total and free testosterone , basal insulin , androstenedione , dehydroepiandrosterone sulfate , 17 - hydroxyprogesterone , thyroid-stimulating hormone , prolactin , follicle stimulating hormone , luteinizing hormone ) and morphological study of the endometrium in both groups. Morphological and immunohistochemical analysis were performed with endometrial biopsies, obtained with video - hysteroscopy. Biopsies were performed in any period of the menstrual cycle in GSOP, because all were in amenorrhea, and in the proliferative phase of the menstrual cycle in GC . Evaluation of endometrial apoptosis for each protein was performed by semiquantitative analysis. Results: The endometria of women with PCOS showed more leukocyte infiltration compared with endometrial of GC. In GSOP, we found significant decrease in immunoreactivity of cleaved caspase-3 and Ki67. No change was observed in Bcl-2, Fas and FasL. Conclusion: Women with PCOS have differences in endometrial apoptosis due to lower expression of caspase-3. The endometrial proliferation index was not increased, in relation to proliferative phase of control patients. Our data suggest that the reduction in apoptosis may be linked to the onset of endometrial diseases.
- ItemSomente MetadadadosEfeito do congelamento sobre o comportamento fenotípico e funcional de linfócitos th1, th2, th17 e treg de indivíduos normais e de pacientes com lúpus eritematoso sistêmico(Universidade Federal de São Paulo (UNIFESP), 2013-10-22) Ferreira, Sanessa da Silva [UNIFESP]; Andrade, Luiz Eduardo Coelho Andrade [UNIFESP]; Universidade Federal de São Paulo (UNIFESP)O Lúpus Eritematoso Sistêmico (LES) é uma doença autoimune crônica, caracterizada por apresentar as mais variadas manifestações clínicas, laboratoriais e imunológicas, cuja etiologia ainda permanece desconhecida. Entre as diversas alterações linfocitárias descritas no LES, destacam-se as alterações de freqüência e função das células Th1,Th2,Th17 e Treg. As respostas destas células T efetoras devem estar em harmonia e o desequilíbrio destas subpopulações possivelmente desempenharm um papel importante na fisiopatologia do LES. Uma maior produção de citocinas de algum desses subtipos de células ou até mesmo a criopreservação pode contribuir para esse desequilíbrio, que pode induzir uma ativação anormal das células T, aumento do processo inflamatório e morte celular induzida por ativação (apoptose). A apoptose é um mecanismo normal de controle de populações linfocitárias, mas quando exacerbado pode levar a maior exposição de autoantígenos e conseqüente desencadeamento e/ou perpetuação de respostas autoimunes. Níveis aumentados de apoptose é um fenômeno comumente observado no LES e pouco ainda se sabe sobre a suscetibilidade das subpopulações Th1,Th2,Th17 e Tregs à apoptose e quais fatores podem atuar para exacerbação do processo. O presente trabalho propõe-se a avaliar o comportamento fenotípico e funcional in vitro das subpopulações Th1,Th2 ,Th17 e Treg sob o efeito do congelamento e descongelamento celular pré-cultivo. Os resultados dessa avaliação deverão contribuir para melhor entendimento de alguns fatores da resposta imune de tais indivíduos, pois verificarão neste trabalho se o fenótipo e a função das células Th1,Th2,Th17 e Treg serão afetados sob diferentes condições de manipulação in vitro.
